Features：Compared to the widely used jRGECOla, FRCaMPi and soma-targeted FRCaMPi (SomaFRCaMPi) exhibit up to 2-fold higher dynamic range and peak ΔF/F0 action potentials in cultured neurons and in vivo. In wide-field imaging of the mouse primary somatosensory and visual cortex with high labeling density (80-90%), SomaFRCaMPi shows up to a 40% improvement in signal-to-noise ratio (SNR) and reduces spurious correlations between neurons. Overall, SomaFRCaMPi significantly enhances the accuracy and scale of neuronal activity imaging at single-neuron resolution in densely labeled brain tissues, with 2 to 3 times improved automated neuronal segmentation efficiency, a 50% increase in the fraction of responsive cells, and up to a 2-fold increase in SNR compared to jRGECOla.
