The U6 promoter is commonly used to drive the expression of small RNAs, such as shRNA or sgRNA. It is a type of RNA polymerase III (Pol III) promoter and is known for its high expression efficiency and strong transcriptional activity.
SasgRNA is designed not only to guide Cas9 to a specific DNA sequence but also to activate gene expression at that location. It does this by incorporating additional sequences that can recruit transcriptional activators, making it capable of inducing gene activation without the need for separate activator proteins. sasgRNA is used primarily in CRISPR activation (CRISPRa) systems where the goal is to upregulate the expression of a target gene rather than edit the DNA.
